You shouldn't see a difference because the optimization has been in for awhile.  The only difference in the new build is the new extension point.  In previous builds the prefixes for all SDK plugins were hard-coded in a file (plugins/org.eclipse.core.boot/classloader.properties).  I seem to recall the improvement was mainly noticed in dev mode (when loading class files instead of JARs).
